چکیده

Reg is a gene associated with regeneration of pancreatic islets. We have previously shown that nicotinamide induces differentiation of human fetal P-cells in tissue culture and that hepatocyte growth factor/scatter factor (HGF/SF) is mitogenic for the fetal p-cells. We now tested whether these conditions, supporting either differentiation or growth, are associated with changes in reg gene expression in human fetal pancreatic cells. Culture for 7 days with 10 mM nicotinamide led to a fourfold decrease in reg mRNA levels (23 ± 12% of control, n = 5, P < 0.001). In contrast, HGF/SF increased reg expression threefold (302 ± 68% of control, n = 4, P < 0.05). Nicotinamide, which does not alter the differentiation level of adult P-cells, did not significantly affect reg expression in adult human islets (84 ± 4% of control, n = 2, NS). Thus, a higher level of endocrine differentiation is associated with a lower level of reg expression, and a higher rate of p-cell replication results in increased reg transcription. These results provide the first evidence of a molecular marker, the reg gene, to distinguish between proliferation and differentiation of human p-cells. Diabetes 43:1164-1166, 1994

Effects of Treatment with Bone Morphogenetic Protein 4 and Co-culture on Expression of Piwil2 Gene in Mouse Differentiated Embryonic Stem Cells

Background Specific growth factors and feeder layers seem to have important roles in in vitro embryonic stem cells (ESCs) differentiation. In this study, the effects of bone morphogenetic protein 4 (BMP4) and mouse embryonic fibroblasts (MEFs) co-culture system on germ cell differentiation from mouse ESCs were studied. MaterialsAndMethods Cell suspension was prepared from one-day-old embryoid bo...

Supernatants From Human Osteosarcoma Cultured Cell Lines Induce Modifications in Growth and Differentiation of THP-1 Cells and Phosphoinositide- Specific Phospholipase C Enzymes

Introduction: Introduction: Molecular components within the microenvironment act upon cell growth, survival/apoptosis, and proliferation. Immune system cells respond to molecules produced by the tumor and released in the surrounding microenvironment, such as cytokines, chemokines, and growth factors. This study aimed to identify the effects of tumor environment on monocyte-macrophage cell linea...

تمایز بن‌یاخته‌های‌ جنینی‌ انسان‌ به‌ سلولهای‌ مولد انسولین‌

Introduction: Type I diabetes mellitus is caused by autoimmune destruction of the insulin-producing β-cells. A new potential method for curing the disease is transplantation of differentiated insulin- secreting cells from human embryonic stem cells. Methods: Human embryonic stem cell lines (Royan H1) were used to produce embryoid bodies. Differentiation carried out by growth factor-mediated se...

Differentiation of Mesenchymal Stem Cell toward the Insulin-like Cells with Lentivirus Vector Mir-375

Background & Objective: Type1 diabetes is characterized by autoimmune destruction of pancreatic β cells, leading to reduced insulin secretion. Differentiation of mesenchymal stem cells (MSCs) into β-like cells offers new ways of diabetes treatment. MSCs can be insulated from the human umbilical cord tissue and differentiate into insulin-producing cells. Material & Methods: Human um...
